Abstract

Many cyclodextrin derivatives have been synthesized to date because they can increase the water solubility of natural cyclodextrins. The modified cyclodextrin as monosubstituted positively charged also has many advantages: increasing the solubility in water, improving chiral selectivity of guest molecules, and enhancing the inclusion complex formation by electrostatic interaction. These derivatives also have a prospect in drug formulations because the product only has one isomer, thus giving a better understanding of drug formulations. This study aims to synthesize and characterize monosubstituted cyclodextrin derivatives, mono-6-deoxy-6-ammonium-β-cyclodextrin chloride, in particular mono-6-deoxy-6-ammonium-β-cyclodextrin chloride (CD-NH3Cl) and mono-6-deoxy-6-(propylammonium)-β-cyclodextrin chloride (CD-PrAmCl). The subject of this research is β-cyclodextrin, while the objects of this research are the synthetic method, characteristics, and physicochemical properties of mono-6-deoxy-6-ammonium-β-cyclodextrin chloride salt. The CD-NH3Cl and CD-PrAmCl can prepared in good yield and can be obtained through several reaction steps, starting with the synthesis of mono-6-O-(p-toluenesulfonyl)-β-cyclodextrin (CD-OTs) and followed by the synthesis of single cationic ammonium cyclodextrin salt obtained through 2 methods, the indirect method for the synthesis of CD-NH3Cl and a direct method for the synthesis of CD-PrAmCl. The CD-OTs was prepared in 2 steps, the synthesis of CD-NH3Cl was obtained in 3 steps, while CD-PrAmCl was synthesized in 2 steps. The products were characterized using UV-Vis, FTIR, 1H, and 13C NMR, and the results showed that the products were successfully synthesized. In addition, this study also carried out the melting and decomposition point test and solubility test in water. The results of the solubility test showed that the solubility of CD-NH3Cl and CD-PrAmCl in water at various temperatures was higher than that of β-CD.
